Answers: A PCI card is a expansion card that fits into a slot that conforms to the PCI specification as used in modern PCs vs the ISA slots used in the original PC and the MCA slots used in the Personal System/2 (PS/2) that IBM initially attempted to replace the original PC specification. These slots allow for a multitude of devices to be added to a computer system thereby expanding it capabilities. With modern PCs, their importance has decreased as most devices can be connected through USB or Firewire. However, many devices function well through PCI slots when they are not meant to be removed on a regular basis, such as internal WIFI and Modems. They also allow expansion of the system bus, so that one can add as much extra USB or even hard drives to the system. Video cards used to use the PCI slots before the faster AGP and now the PCI-Express (PCIe) slots were introduced.

Can it be used with your system, if you have slots available... yes, e-Machines did not exist in the ISA or MCA days, so most likely it would be a PCI bus machine and therefore can handle installation of new devices that fit the PCI specification, this however does not say if you system can handle newer devices as there is also a limits by older BIOS and the Operating System used.
